Human embryonic stem cells

How do we characterize our human embryonic stem cell lines?

Human embryonic stem cells are characterized as such by a batch of tests. In our lab, these include:

  • Immunostaining for SSEA-3, SSEA-4, TRA-1-60 and TRA-1-81
  • Gene expression analysis by RT-PCR for POU5F1, NANOG, LIN 28, REX1, NPM1, SOX2, DNMT3B and GDF3
  • Alkaline phosphatase activity test
  • Generation of teratomas containing the three germ layers in SCID mice
  • Karyotyping by G-banding or array-based comparative genomic hybridization
Immunostaining for SSEA3 and SSEA4
Immunostaining for TRA-1-60 and TRA-1-81
Gene expression analysis by RT-PCR
Teratoma formation
Karyotype by G-banding
